Fluorogen activating and shifting tags (FAST) are a new family of fluorescent proteins, engineered from the photoactive yellow protein (PYP), that enable the fluorescent labeling of polypeptides/proteins of interest in vitro and in vivo. Their fluorescent properties arise from the reversible and selective binding of a fluorogenic chromophore, which is almost non-fluorescent when free in solution but becomes strongly fluorescent when bound in the protein. Binding of the chromophore is accompanied with a large red-shift of the absorption properties of the chromophore enabling selective excitation of FAST. The reversible binding of the fluorogenic chromophore displays fast dynamics, which enables a permanent renewal of the chromophore in the protein tag and provides FAST with an apparent photostability.
Possible Applications:
Use for quantification and detection of any polypeptide of interest in any solid or liquid sample, as well as in any cell or tissues or organisms of interest using fluorescence as read-out : molecular biology, cell biology, developmental biology, neurobiology, immunology and physiology, biotechnology, biological research, agriculture.
